Script used here - quantiflash_analysis.R

Data/QF scripter - TBU

Note -

  • Scattered signals are not displayed as they are irrelevant. Quantiflash does not emit well <400nm hence BUV395 plot is N/A.
  • Recorded .fcs files need to be in a subfolder called 'laser_on'.
  • With this data - an optimal range of voltages can be determined. Next update will provide for minimal voltage where S/N plateaus as well as numerical deviation from linearity of the detector.

Protocol - 

  1. New experiment, all lasers on. 
  2. Set laser delays to 0
  3. Set all scaling factors to 0.5.
  4. Acquire data with QF scripter automatically changing the QF intensity every second. 
  5. Set FSC so that all H/W FSC signals are on scale. 
  6. Set events to record to 200k.
  7. Record data with PMT volts from 0-1000 (25V increments) - PM for app to do this unattended. 
  8. Put data in sub folder labelled 'laser_on' and run R script.
