Quantiflash Data - Symphony
Note -
- Scattered signals are not displayed as they are irrelevant. Quantiflash does not emit well <400nm hence BUV395 plot is N/A & in the NIR hence why these plots look odd.
- Recorded .fcs files need to be in a subfolder called 'laser_on'
- Recorded .fcs files need to be in a subfolder called 'laser_off'
- With this data - an optimal range of voltages can be determined.Â
- Height parameter used - has in testing matched manual voltration in testing.Â
Protocol -Â
- New experiment, all lasers on.Â
- Set laser delays to 0
- Set all scaling factors to 0.5 (inc. FSC)
- Acquire data with QF scripter automatically changing the QF intensity every second - 1000 events/second, gaussian pulse (5 sec) - standard settings (FSC set to 400V), record 20k events. Record all HWA signals on all channels, FL in LOG.Â
- Set FSC so that all H/W FSC signals are on scale.Â
- Set events to record to 20k.
- Record data with PMT volts from 0-1000 (25V increments) - PM for app to do this unattended (vchanger.exe)
- Put data in sub folder labelled 'laser_on' and 'laser_off' and run R script - request via email.Â
