Note -
- Scattered signals are not displayed as they are irrelevant. Quantiflash does not emit well <400nm hence BUV395 plot is N/A.
- Recorded .fcs files need to be in a subfolder called 'laser_on'.
- With this data - an optimal range of voltages can be determined. Next update will provide for minimal voltage where S/N plateaus as well as numerical deviation from linearity of the detector.
- Height parameter used.Â
Protocol -Â
- New experiment, all lasers on.Â
- Set laser delays to 0
- Set all scaling factors to 0.5.
- Acquire data with QF scripter automatically changing the QF intensity every second.Â
- Set FSC so that all H/W FSC signals are on scale.Â
- Set events to record to 200k.
- Record data with PMT volts from 0-1000 (25V increments) - PM for app to do this unattended.Â
- Put data in sub folder labelled 'laser_on' and run R script.
