Cryo fixation - high pressure freezing and freeze substitution

High pressure freezing is used to freeze biological samples in a vitreous state, preventing the formation of any detectable ice crystals. This is achieved by rapidly cooling the sample with liquid nitrogen while simultaneously exposing it to very high pressures (2100 bar). The sample is then transferred to a freeze substitution unit where it is fixed, dehydrated and processed to a resin block at low temperature, preventing the typical processing artefacts that are introduced during conventional room temperature processing. High pressure freezing and freeze substitution gives superior ultrastructural cellular morphology to room temperature processing.
